ULTRAFAST METHOD FOR SIMULTANEOUS MEASUREMENT OF TRIAZOLE ANTIFUNGALS IN HUMAN SERUM USING ONLINE SP

Abstract

Clinical research laboratories traditionally rely on HPLC and LC/MS/MS for quantitative analysis of antifungal drugs. This Application Note describes a rapid and robust online SPE/MS/MS analytical method for high-through put and accurate measurement of five antifungal drugs (voriconazole, ketoconazole,fluconazole, itraconazole, and posaconazole) in human serum. This methodemploys protein precipitation followed by dilute and shoot analysis on a SPE/MS/MS system, enabling the analysis of all five antifungals at a rate of 14 seconds per sample. This analytical method produces > 10x savings in analysis time and solvent consumption compared to typical HPLC-based methods.

Abstract

The measurement of gabapentin and pregabalin in urine requires a reliable, efficient, and accurate method. We developed an analytical method using a SPE/MS/MS system to measure these analytes in urine with much faster sample cycle times, and similar analytes in urine with much faster sample cycle times, and similar analytical results compared to traditional methods. A simple dilute-and-shoot based analysis by the Agilent RapidFire/MS/MS permits accurate and precise measurement of these analytes over a linear range of 0.5 to 100 mcg/mL. Samples were analyzed at 25 seconds per sample, providing a much higher throughput method of anlysis compared to traditional protocols.

Ultrafast Analysis of Clozapine and Norclozapine in Serum Using the Agilent RapidFire High-Throughput Mass Spectrometry System

The steadily increasing need for greater analytical capacity and throughput have placed high demands on traditional technologies in the clinical research laboratory. The Agilent RapidFire High-throughput Mass Spectrometry System is an ultrafast SPE/MS/MS system capable of analyzing samples with cycle times less than 15 seconds. The quantitative capability of the Agilent RapidFire/MS/MS system to analyze clozapine and its metabolite, norclozapine, was evaluated in human serum. This analytical method employs protein precipitation followed by dilute and shoot on the SPE-MS/MS system, enabling analysis of these analytes at 14.5 seconds per sample, producing > 10x savings in analysis time and solvent consumption compared to typical LC/MS/MS methods.

Abstract

Ultrafast Analysis of a Tricyclic Antidepressant Drug Panel in Human Serum by the Agilent RapidFire High-Throughput Triple Quadrupole Mass Spectrometry System 

Quantitative analysis of tricyclic antidepressant drugs (TCAs) in clinical research laboratories traditionally relies on HPLC and immunoassay, however interfering substances, false positives and cross reactivity to other cyclic compounds may compromise clinical research results.  An efficient, fast, accurate, and sensitive SPE/MS/MS method with a wide calibration range was developed for the simultaneous quantitation of 8 antidepressant drugs in human serum (Amitriptyline, Nortriptyline, Imipramine, Desipramine, Doxepin, Nordoxepin, Clomipramine, and Norclomipramine). This method employs protein precipitation followed by dilute and shoot on the SPE/MS/MS system, enabling analysis of all 8 TCAs at 13 seconds per sample producing >10x savings in analysis time and solvent consumption compared to typical HPLC methods.

Abstract

Ultrafast Analysis of Z-Drugs in Urine Using the Agilent RapidFire High-Throughput Mass Spectrometry System

An ultrafast RapidFire/MS/MS method was developed for simultaneous determination in human urine of sleep aids (Z-drugs): Zopiclone, Zolpidem, and Zaleplon.  The need for greater analytical capacity and throughput for the analysis of sleep aid medicines (Z-drugs) in forensic toxicology laboratories has placed demands on traditional analytical technologies – GC/MS and LC/MS/MS. We developed a method utilizing the Agilent RapidFire/MS/MS system to analyze Z-drugs in urine with much faster sample cycle times and similar analytical results compared to LC/MS/MS assays. A simple dilute and shoot methodology followed by analysis by RapidFire/MS/MS allows for the accurate and precise measurement of these analytes in urine over a linear range of 5 to 500 ng/mL. Samples were analyzed on the RapidFire/MS/MS system at 15 seconds per sample providing a much higher throughput method of analysis compared to traditional LC/MS/MS protocols. This new ultrafast method has the speed and accuracy necessary for an efficient screen/confirm (qualitative and quantitative) workflow.

This method employs protein precipitation followed by dilute and shoot on the SPE/MS/MS system, enabling analysis of all 6 AEDs at 14 seconds per sample producing >10x savings in analysis time and solvent consumption compared to typical LC/MS/MS methods.
